Effect of Methotrexate on the Liver of Rats

Abstract
In order to study the effect of methotrexate on the liver functional tests and histochemical studies on the liver were carried out after intramuscular administration of methotrexate to the rats. The results were as follows.
1. In the group of short(2 five-day) course administration, the serum levels of. glutamic pyruvic transaminase,and alkaline phosphatase increased slightly. In the long(4 five-day) course group, thymol turbidity and serum alkaline phosphatase level increased to slight to moderate degree.
2. The hepatic changes in the short course group were focal necrosis of fused liver cells, intracytoplasmic granular deposition of lipid, thrombosis in the portal and central veins. But inflammatory reaction was not observed. In the long course group, however, inflammatory cell infiltration which was consisted chiefly neutrophils and lymphocytes as well as more conspicuous necrotic changes were revealed. Other- hepatic changes involed Kupffer cell proliferation, presence of acidophilic hyaline bodies in the.liver cells, cystic dilatation of sinusoids in the central zone and globular fatty deposition in the liver cells.
3. Reticular fibers, on the reticulum stain, disrupted, condensed or even disappeared in the areas of patchy cellular mass and cystic dilatation.
4. PAS, adenosine triphosphatase and alkaline phosphatase reactions showed no significant changes in both groups of short and long course administration.
